MT concentrations in both organs were found to be lower than normal values at all time points. Maximum levels were reached after 10 days of administration at a dose of 5 mg Cd/l and 15 mg Zn/l. As all MT concentrations in liver tissue were below normal values already after 5 days of administration, we conclude that this organ is more sensitive to the toxic effects of Cd and Zn than kidney. In liver there was a positive correlation of the MT concentration with the glutathione (GSH) concentration. After cessation of the treatment the MT concentration and GSH concentration were found to return to normal levels within a short period of 2-3 weeks. In kidney, however, a normal MT concentration was only reached after 7 weeks with a dose of 5 mg Cd/l and 15 mg Zn/l. In Zn-administrated animals the GSH concentration remained at normal values even after 22 days. During administration of Cd, the GSH concentration was found to decrease continuously and the decrease of the GSH concentration lasted even longer than after administration of Zn. At the lowest dose Cd treatment leads to a linear decrease of the GSH concentration, but at the higher dose, GSH levels remain within the control range. The increase of the GSH concentration in Zn-administered rats is mainly a result of an increase in the glutathione-S-transferase activity. In kidney, the fall in GSH concentrations can be explained by the decrease of the activity of the glutathione-S-transferase in the tubular cell membrane.
