Recently, we have shown that insulin activates the mitogen-activated protein kinase/extracellular signal-regulated kinase (MAPK/ERK) pathway in mesangial cells (MCs) via dendrin-induced activation of phosphoinositide 3-kinase (PI3K). Here, we show that Akt/protein kinase B (PKB) also acts as a critical signaling molecule in insulin-induced MC differentiation. Simultaneous activation of PI3K and Akt/PKB is required for insulin-induced MC differentiation, and Akt/PKB activation is mediated by dendrin but not by IGF-1. Because p85beta, but not p85alpha, plays a critical role in the activation of Akt/PKB by dendrin, the molecular link between dendrin and p85beta should be explored in detail. In addition, we found that insulin induced Akt/ERK pathway activation through MEK1/MEK2. Interestingly, cells expressing dominant-negative dendrin, which inhibits the Akt/ERK pathway, are unable to differentiate in response to insulin.
